Is tuning via a dyno essential subsequent to the install of a supercharger, or in the event that one does not have access to a dyno, would a specific tune from a vendor via an Xcal2 suffice in order to run the car safely? Also, does anyone have any opinions on intercooled vs. non intercooled set ups as far as durability is concerned?

I think the tune provided with any of the S/C kits should be fine until you can get to a tuner.
I believe RF stated the tune he relieved with his kit turned out to be spot on after getting to a Dyno.

I would at least get a good wideband O2 setup to verify a healthy air to fuel ratio, which is one of the main goals on a dyno anyway.

Intercooled reduces the charged temp allowing more boost/spark/hp.

It does not affect durability.

~450RWHP on a Dynojet appears to be the "sweet spot" for durability for a DD car.

There are a number of S/C w/ 40K-80K miles ~450RWHP and no problems. There are several w/ ~500RWHP that blew up. There are several ~500RWHP garage queens that have not blown up yet.

My s/c kit did NOT run well w/ the provided Tune. The new MAF was not calibrated (I have been told this is normal w/ an SCT2800).

I believe RF stated the tune he received with his kit turned out to be spot on after getting to a Dyno.RC is correct. I had Lidio dyno mine about a month after the install (done locally) and he said it only needed a minor tweak to the A/F.

What is the optimum air/fuel ratio? I assume that in the event an adjustment is required, that some tweaking can be done via the xCAL2.

Personally... I would not touch A/F w/o a Dyno, or Wideband. I don't care WHAT the XCal2 CAN do.

If one is able to get the A/F to a point utilizing a wideband and Xcal2, is that it? By that, I mean has a person set the parameters without future need for adjustments dependent upon temperature or other factors? Perhaps a strange question to those in the know, but I just want to make certain that I understand correctly. It would also seem then, that an A/F gauge would not be a prerequisite, but perhaps just convenient to have.
